FAQ: What is more sensitive – measuring from the top or bottom of a plate?
A top read measurement (with the lid off) will always give a more sensitive reading than a bottom read measurement for two reasons:
Most instruments use fiber optics to read from the bottom of the plate, which lose light over distance.
The plastic bottom of the microplate will absorb light before it is measured, thereby decreasing the signal further.
